Relief of Pain and Inflammation : Gout
OCOLC1 “Colchicine tablet” 0.5 mg/tab
適應症:痛風。
Usual dose:
Acute gout:
Initially 0.5-1.2 mg, then 0.5-0.6 mg q1h or 1-1.2 mg q2h until symptoms abate or diarrhea occurs (usual dose 4-8 mg); max. 8 mg per attack; a 3-day colchicine-free interval should follow each oral course.
Gout; Prophylaxis:
< 1 acute attack/yr, 0.5-0.6 mg/day 3-4 times/wk;
> 1 acute attack/yr, 0.5-0.6 mg/day up to 1.5-1.8 mg/day.
Dose adjustment:
Hepatic insufficiency: dose reduced, careful monitoring.
Renal impairment: CrCl: 10-50 ml/min, < 0.6 mg PO daily or qod; CrCl < 10 ml/min, contraindicated.
Contraindication:
Blood dyscrasias, cardiac disease, gastrointestinal disease, hepatic failure, hypersensitivity to colchicine, renal disease.
Adverse effect:
Common: diarrhea, epigastric pain, nausea, vomiting.
Serious: myelosuppression, neuromyopathy.
