適應症：圓盤狀及全身性紅斑性狼瘡、慢性多形日光疹、慢性風濕性關節炎、鏈狀瘧原蟲和間日瘧原蟲引起之瘧疾。
Usual dose:
Malaria, acute attack:
Adult: 800 mg initially, then 400 mg in 6-8 h and 400 mg qd for 2 days or a single dose of 800 mg.
Pediatric: initially, 10 mg base/kg, max. 620 mg base (1st dose), 5 mg base/kg in 6 h, max. 310 mg base (2nd dose), then 18 h after 2nd dose (3rd dose), then 24 h after 3rd dose (4th dose).
Rheumatoid arthritis:
Adult: initially, 400-600 mg qd for 4-12 wks; maintenance, 200-400 mg qd.
Systemic lupus erythematosus:
Adult: initially, 400 mg qd or bid until remission; maintenance, 200-400 mg qd.
Adverse effect:
Common: drug-induced pigmentation, nausea, vomiting, diarrhea, drug-induced myopathy, headache.
Serious:agranulocytosis, disorder of eye, ototoxicity.
