Antineoplastic Agents: Cytotoxic Agents: Alkylating Agents

適應症：多發性骨髓瘤．卵巢癌．真性紅血球過多症。
Usual dose:
Epithelial ovarian cancer: PO, 0.2 mg/kg/day for 5 days; repeat q4-5w depending on hematologic tolerance.
Multiple myeloma (monotherapy): PO, 6 mg/day for 2-3 wks, then 4 wks off, then when blood cell/platelet counts are rising 2 mg/day; other regimens include 0.25 mg/kg/day for 4 consecutive days (0.2 mg/kg/day for 5 days) in combination with prednisone- total dose of 1 mg/kg/course repeated q4-6w if granulocyte/platelet counts have returned to normal; 10 mg/day for 7-10 days with a maintenance dose of 2 mg/day; 0.15 mg/kg/day for 7 days with a maintenance dose of 0.05 mg/kg/day or less; (combination therapy): melphalan 10 mg/m2/day PO with prednisone 60 mg/m2/day PO 1-4 repeat cycle q42d.
Adverse effect:
Common: alopecia, stomatitis.
Serious: jaundice, hepatitis, hemolytic anemia, myelosuppression, pulmonary infiltrates/fibrosis, secondary malignancies, vasculitis.
