Anti-infective Agents : Antiretroviral Agents
O3TC1 “3TC FC tablet” 150 mg/tab
適應症：與其他抗反轉錄病毒劑合併使用，治療成人及兒童之人體免疫缺乏病毒(HIV)感染。
Usual dose:
HIV infection, in combination with other antiretrovirals:
Adult: 150 mg bid or 300 mg qd.
Pediatric
(3 mon-16 yrs): 4 mg/kg bid, max. 150 mg bid.
(neonates, < 30 days): 2 mg/kg bid.
Perinatal exposure; prophylaxis: 150 mg plus zidovudine 600 mg to the mother at onset of labor, followed by lamivudine 150 mg q12h, zidovudine 300 mg q3h until delivery; postpartum the neonate should receive lamivudine 2 mg/kg plus zidovudine 4 mg/kg q12h for 7 days.
Dose adjustment:
Renal failure: (HIV infection) CrCl 30-49 ml/min, 150 mg qd; CrCl 15-29 ml/min, 150 mg first dose then 100 mg qd; CrCl 5-14 ml/min, 150 mg first dose then 50 mg qd; CrCl < 5 ml/min, 50 mg first dose then 25 mg qd.
Adverse effect:
Common: lipodystrophy, decrease in appetite, nausea and vomiting, headache, fatigue.
Serious: lactic acidosis, pancreatitis (especially children), hepatomegaly, relapsing type B viral hepatitis.
